Ribo-MET: Detection of DNA Methylation Ribo-MET is a product line for the detection of changes in methylation levels of genes and histones. Methyl groups attached to certain amino acids in histone proteins change the accessibility of the DNA to the proteins that turn on genes. Methyl molecules attached to certain regulatory regions of the DNA determine whether or not a gene is turned on or “expressed”. Increased methylation of a DNA sequence is usually associated with gene silencing. The control of gene expression by methylation plays a critical role in maintaining normal cell function by turning genes on and off at the appropriate times. Too much methylation or too little methylation can cause disease, including cancer.

Ribomed’s first methylation tests, Ribo-MET are for detecting CpG methylation of DNA. Ribo-MET tests are designed to work on both native DNA and DNA that has been chemically modified with bisulfite. Ribo-MET tests are being formatted for detection of a few targets in a sample using extremely inexpensive instrumentation for the basic research market or for detection of multiple targets and high throughput screening by the pharmaceutical industry. The first Ribo-MET kits for the research market should be completed and ready for beta testing in 2008. Ribomed is currently selling several of the individual components of these tests.

Ribo-miRNA: Detection of Micro RNAs Ribo-miRNA kits are for the detection of multiple small RNA molecules involved in the inhibition of gene expression and associated with the onset of disease. Unlike mRNA, miRNA does not code for protein production. Instead, it acts by binding to mRNA and changes its ability to be used as a template for protein production. There are hundreds of miRNAs in the cell that are believed to play regulatory roles in many processes, including those involved in cancer, viral disease, diabetes, leukemia and neurodevelopment.

Ribomed’s Ribo-miRNA will be radically different than any existing detection technology. Abscription is uniquely suited for the detection of these very small RNA molecules and our Ribo-RNA tests have already been validated for sensitivity and better specificity than RT-PCR during our development of detection tests for RNA viruses for the government. RT-PCR is the method most commonly being used for miRNA detection, and it is a difficult process with such a small RNA target.
